Can Troglodytia still use her ability after the very same player has Suddened the master card she wants to use her speeciality on?
Player A: I play Villein.
Player B: I play Sudden Reversal on Villein.
Player B: I use Troglodytia's special ability on the Villein.
Should you actually use her specialty BEFORE playing the Sudden Reversal?
I figured that since Sudden was played (by the same player no less), the window for Troglodytia's special ability had already gone.

Independent: After any Methuselah plays a master card, you may look at that Methuselah's hand (after the replacement card is drawn). +1 strength.
Sudden Reversal
Cancel a master card played by another Methuselah as it is played (no cost is paid).

"as it is played" occurs even before the card is replaced, so there's no doubt here.
A plays Villein (not replaced)
B plays Sudden Reversal (not replaced)
B replaces SR
A replaces Villein

Yeah, indeed you need to use Sudden before replacements are drawn.
People tend not to play that way and we all had a communal lapse of thought regarding that.

Is "after" before or after the master cards effect? Like when you use Troglodytia's special to Blessing of the Beast, do you look the hand before or after you put Spiritus cards to BotB?

I can't find a precedent ruling for this.
So we'll go with what already exists, and that would be cardtext (in our case, Permanency).
"After X is played" is after resolution of X.
Regarding Blessing of the Beast vs Troglodytia: You look at the hand after the Spiritus cards were placed on the Blessing of the Beast, and after they were replaced.
