Card effects that damage the vampire may be used, yes. If the damage is not
aggravated, however, it will be ignored.

Yes, you can. The aggravated damage will affect the Nihilistic vampire
as usual, since Ex Nihilo doesn't have anything to say about aggravated
damage.
While the official card name is 'Rutor's Hand,' there is something rather
charming about 'Rupture's Hand,' if that is what you intended to write.

That is precisely correct, if you mean the damage will be ignored. Other,
non-nothing things will continue to happen, however.

Damage is only considered to have come from a minion if the damage is from a
strike or if an effect specifically states that the damage is dealt by the
minion. Damage from other effects is considered "environmental," and has no
particular source.

Wolf Companions, Zombies, Murders of Crows, Ghoul Retainers, Weather Control,
and Carrion Crows all inflict damage as normal. Strikes from the
Nihilistic vampire (including superior Stutter-Step and Catatonic Fear, as
well as Riposte, since these cards use the word 'inflict') do no damage.
Useful strikes under such conditions include Flesh and Bonecraft (no damage,
but side-effects apply), Weakness (note that the vampire takes 1 damage at
superior, since the passive 'takes one damage' is used, indicating an
environmental effect), Entombment, Coma, steal/destroy equipment (note
that superior Fractured Armament still inflicts damage), and steal blood.
