To expand on this question a bit: If opposing vamp's controller
"jumps the gun" and attempts to play Rotschreck immediately after
anouncing her strike and then the D.T. is played will it cancel the
Rotschreck or force the Methuselah to put it back in her hand? The
answer to that should answer this question but I will ask anyway, If
Marconius has some kind of damage prevention (Flak Jacket, Guardian
Angel) and does not care to use the D.T. at the moment and then the
opposing vamp's controller plays Rotschreck can he respond by playing
the D.T. since both happen before strike resolution?

There's no "jumping the gun" allowed - R. and DT have the same timing,
so the acting Methuselah gets first chance to play.
If he plays DT, then R. cannot be played.
If he passes and then his opponent plays R., then DT cannot be played.

Would Skin of night have the same effect, played before strike
resolution? eg Agg damage trated as regular damage.
Does R ignore this as it's this vampire with SoN and the opposing
vamp is still trying to do agg damage?

Right, SoN doesn't change the damage being inflicted, so Rotshreck can
still be played.

No. SoN doesn't alter the fact that the infliction of aggravated damage
(1 or more points) is being attempted.
