A vampire must be untapped to use a reaction card, yes.

You can give up the edge whenever you want to, including once it's "clear"
that a bleed will be successful. Note that it's possible that you would
think you're about to bleed, so you give up the edge, so your opponent
Telepathic Counters the bleed away.

It would be after the combat.

You spend one master action to take one counter off of one Investment.
You can't use multiple Investments unless you have multiple master actions
to spend.
